NAD+ Precursors Compared: NMN vs NR vs Niacin vs NAM — The Complete 2026 Guide
If you’ve spent any time in the longevity world, you’ve heard that NAD+ is critical for aging. Older organisms have lower NAD+. NAD+-restoring interventions extend lifespan in animals. Therefore, you should take NAD+ precursors to stay young. The logic feels airtight. But it’s wrong—or at least, it’s incomplete.
NAD+ is not a vitamin you can simply “take.” It’s a molecule your cells make from precursors, and how you get those precursors to the right place, in the right form, at the right concentration, determines whether you see any benefit.
The Four Main Precursors: What Each Does
Nicotinamide Riboside (NR)
NR is a nucleoside that’s been aggressively marketed as a longevity supplement. The animal evidence is real. In yeast, worms, and mice, NR administration increases NAD+ levels and improves multiple aging phenotypes.
The human evidence is weaker. We have short-term studies showing that NR increases NAD+ in blood and tissues. We have no long-term, large-scale human trials showing lifespan or healthspan benefits.
Nicotinamide Mononucleotide (NMN)
NMN is one step closer to NAD+ than NR. The animal evidence for NMN is stronger than for NR. In mice, NMN increases NAD+, improves physical function, enhances mitochondrial biogenesis, and extends lifespan.
The human evidence is growing but still limited. We have small studies showing that NMN increases NAD+ and improves muscle strength in aging populations.
Niacin (Nicotinic Acid)
Niacin is vitamin B3. Your body converts niacin → nicotinic acid mononucleotide → NAD+. The advantages are obvious: niacin is cheap, widely available, and has a long safety record. The disadvantage is the niacin flush—vasodilation causing heat, redness, and itching.
Nicotinamide (NAM)
Nicotinamide is another form of vitamin B3. The problem is that nicotinamide is a SIRT1 inhibitor. By inhibiting SIRT1, nicotinamide might counteract the anti-aging effects of NAD+. I don’t recommend nicotinamide for longevity purposes.
Final Verdict
For most longevity-focused adults, NMN edges out NR because of superior animal data. For cost-conscious individuals, niacin is an excellent alternative. Do not take nicotinamide.
